Magaldrate is an antacid have been widely used in the treatment of various gastric and duodenal disorders such as heartburn, reflux esophagitis, acid indigestion, gastritis, sour stomach, upset irritable ulcers. The conventional antacids dosage forms a short duration action which about 2-3 hours due to emptying process. A gastroretentive form needed since healing ulcers occurs when pH kept above 3-4 during 24 hours. present study was aimed at developing Gastro retentive bilayer drug delivery systems containing minimize side effect, improve prolongation action, reduce frequency administration. wet granulation technique formulate 9 batches. Superdisintegrants like Polyplasdone XL-10, Ac-Di-Sol, sodium starch glycolate for immediate release layer HPMC K4 M, Ac-Di-Sol lactose polymers were floating layer. Preformulation studies carried out optimize ratios required grades polymers. prepared tablets evaluated hardness, weight variation, thickness, friability, content uniformity, buoyancy lag time, total water uptake (swelling index), vitro dissolution studies. Successful formulation developed having time low 30 sec sustained up 12 hrs. biphasic can be obtained by using tableting technology involved compression together. Bilayered with characteristics offer critical advantages as, site specificity improved absorption efficacy. This inculcated medicaments stomach major absorption.
